Cognitive flexibility requires rapid updating of internal representations when action–outcome contingencies change. Yet how feedback on a single trial reshapes the distributed activity patterns that implement these internal representations remains poorly understood. Using functional magnetic resonance imaging (fMRI) during probabilistic reversal learning, we show that a Q-learning model with separate learning rates outperforms a standard model and reveals stronger learning from positive than negative reward prediction errors (RPE). Reversal increases prediction error magnitude and induces rapid multivoxel reorganization in medial temporal and adjacent subcortical regions. During early reversal, RPE-related activity in right striatal and medial orbitofrontal regions predicts subsequent changes in right lateral orbitofrontal cortex (lOFC) representations; these effects occur only after nonrewarded outcomes. Moreover, greater representational change in lOFC is associated with weakening of the action policy, and this pattern change in right lOFC is related to reward-stay behavior. Together, these findings link lOFC updating to behavioral policy adjustment during reversal learning.

How is vision shaped by other senses? Visual cortex is not merely a passive recipient of retinal inputs. It is also responsive to other sensory modalities, particularly audition. While auditory signals have been shown to penetrate early visual cortices, the mechanisms driving these responses remain poorly understood. To examine how audition shapes vision, we used functional MRI to investigate whether audiovisual interactions modulate the gain of responses in human early visual cortices. Specifically, we measured voxel-wise contrast-response functions (CRF), while participants viewed horizontally drifting gratings paired with auditory motion that was directionally congruent or incongruent with the visual motion, or stationary. We show that congruent audiovisual motion enhances voxel-wise contrast sensitivity whereas incongruent motion reduces it, manifesting as a robust contrast gain modulation across areas V1–V3. This direction-selective enhancement is nicely captured by a divisive normalization model incorporating auditory-driven feedback modulation. Our findings provide compelling evidence for cross-modal interactions at early stages of visual processing, driven by contrast gain modulation –a bidirectional modulation in sensitivity that maintains the balance between local excitatory and inhibitory activity. Together, these results reveal that audiovisual interactions can alter a cornerstone property of vision, highlighting the dynamic role of early visual cortex in multisensory processing.

Sleep has been linked to brain waste clearance and glymphatic/perivascular transport, yet whether insomnia shares genetic architecture with perivascular diffusivity remains unclear. Here we show that insomnia liability and diffusion tensor imaging measures along the perivascular space have little genome-wide genetic correlation but converge locally at chromosome 17q21.31. Polygenic enrichment, conjunctional false discovery rate, and Bayesian colocalization analyses support shared statistical signals in this region, which contains the MAPT inversion locus. Transcriptome-wide, gene-level fine-mapping, and summary-data Mendelian randomization analyses prioritize HEXIM1, ACBD4, EFTUD2, and MAPT. Tissue, cell-type, spatial transcriptomic, and sleep-perturbation analyses place these candidates in neuronal, glial, immune, vascular, and perivascular contexts, with effects varying by brain region and imaging phenotype. Together, these findings support localized genetic convergence rather than a genome-wide shared architecture and provide biological context for future studies of perivascular imaging phenotypes and neurodegeneration-related processes. Insomnia and MRI-derived perivascular diffusivity show little genome-wide genetic correlation but converge locally at 17q21.31, highlighting HEXIM1, ACBD4, EFTUD2 and MAPT.
